地黄愈伤组织诱导条件的研究

摘    要:为寻求诱导地黄愈伤组织的最佳培养条件和激素配比,研究了地黄不同外植体、激素浓度及其组合、培养基种类等条件下暗培养对地黄愈伤组织诱导的影响。结果表明:以地黄叶片、叶柄、茎段作为外植体在MS+1.0~1.5 mg/LNAA条件下诱导率均能达到100%,其中叶片诱导的愈伤生长势较好;单独使用NAA和2,4-D均能高频率诱导出愈伤,加入0.5 mg/L6-BA后明显提高生长速度;MS、LS培养基诱导率高,但B5培养基中愈伤生长势较好。

Abstract:To gain the optimization conditions of the callus induction,the research on the inducement conditions of the callus from the different tobacco explants was conducted and the different culture media were added with various concentrations of 2.4-D,NAA and 6-BA.The result indicated that the rate of callus induction from leaf,petiole and stem segment was 100% on the medium with MS + 1.0~1.5 mg/L NAA,but more callus from leaf was inducted.Two kinds of auxins could induce callus,but the callus grew faster in the medium added with 0.5mg/L 6BA.MS and LS medium was more suitable for the callus induction and the callus from B5 medium grew best.
