摘    要:目的]开发新鲜蔬菜中有机磷农药残留的快速检测技术。方法]根据有机磷农药抑制胆碱酯酶活性的原理,开发了一种快速检测新鲜蔬菜中有机磷农药残留的技术,由复合指示剂颜色变化和反应液△pH值进行定量,通过单因素试验和正交试验确定最佳反应条件。结果]复合指示剂的配比为溴百里香酚蓝钠盐∶石蕊指示剂=2∶3。马血清的添加量对△pH值的影响最大,其次为氯化乙酰胆碱浓度,温度的影响最小。最佳反应条件为:马血清0.8 ml,1.0%氯化乙酰胆碱,温度33℃,该条件下,敌敌畏、辛硫磷、敌百虫的最低检出限分别为0.06、1.00、0.50 mg/kg。结论]该农药残留检测技术方便、快速,在有机磷农药残留的快速测定中有广泛的应用价值。

Abstract:Objective] The research aimed to develop the rapid detection technology of organophosphorus pesticides residue in fresh vegetables.Method] A rapid detection technology of organophosphorus pesticide residue in fresh vegetables was developed according to the principle of organophosphorus pesticides inhibiting the cholinesterase activities and quantified by the color change of complex indicator and the △pH of reaction solution.The optimum reaction conditions were determined by the single factor experiment and orthogonal test.Result] The ratio of complex indicator was as follows: the sodium salt of bromthymol blue∶litmus indicator=2∶3.The addition of horse serum had highest effect on △pH,followed by the concn.of acetylcholine,and the temperature had lowest effect on △pH. The optimum reaction conditions were as follows: 0.8 ml horse serum,1.0% acetylchdinesterase and the temperature of 33 ℃.Under these conditions,the lowest detection limit of dichlorvos,phoxim,dipterex was 0.06,1.00 and 0.50 mg/kg,resp.Conclusion] The detection technology of pesticides residue was convenient and rapid,and it had extensive application value on the rapid detection of organophosphorus pesticides residue.
