罗非鱼鱼皮酸溶性胶原蛋白的制备工艺研究

摘    要:目的]研究从罗非鱼鱼皮中用酸溶剂提取胶原蛋白的最佳条件.方法]以罗非鱼鱼皮为原料,利用酸处理提取胶原蛋白.以提取溶液、浓度、浸提时间、料液比为因素,通过测定其羟脯氨酸含量,计算不同提取条件下胶原蛋白的得率.再利用响应面法对提取条件进行优化,对拟合多项式回归模型进行分析.结果]由响应面优化得到罗非鱼鱼皮中胶原蛋白最佳酸提取工艺条件为:乙酸浓度0.53 mol/L,浸泡时间64.8h、料液比1∶25 g/ml,此条件下胶原蛋白提取量试验均值为244.86 mg/g.结论]研究可为罗非鱼鱼皮中胶原蛋白的提取利用提供参考.

Study on the Extraction of Acid-solubl College from Oreochromis nilofscus Skin

Abstract: Objective ] To study the optimum conditions of extracting collagen using acid solution from the skin of Tilapia. Method ] The ex- tracting technology of collagen using acid solution from the skin of Tilapia Was studied in this paper, Choosing acid type, concentration of acid solution extraction time and solid-liquid ratio as single factor experiment factors, :the extracting results were achieved by measuring the hydroxyproline content, the yield of collagen under different extraction conditions were calculated. The optimal extracting conditions were in- vestigated based on box-behnken design and response surface methodology. Result] By analyzing the fitted polynomial regression model, the optimum parameters of extracting collagen from the skin of Tilapia were as follows : soaking Tilapia skin in acetic acid solution 0.53 mol/L with solid-liquid ratio 1:25 g/ml at 4 ℃ for 64.8 h, and the optimized extraction led to a collagen content of 244.86 mg/g. Conclusion] The study will provide reference for extraction and utilization of collagen from skin of Tilapia.
