紫花含笑的组织培养技术研究

摘    要:[目的]为紫花含笑的开发和大量繁殖提供参考。[方法]以紫花含笑成年树带芽茎段为材料,预处理后接种到诱导培养基上诱导不定芽,然后分别对不定芽进行增殖和生根培养,研究不同外植体处理方法、培养基、激素种类及配比对紫花含笑培养效果的影响。[结果]4 ℃低温预处理5~7 d可降低外植体的褐化及污染率;紫花含笑的最佳基本培养基为MS培养基,激素配比为6 BA 0.5 mol/L+IBA 0.1 mol/L,继代周期以25 d为宜,增殖系数可达3.5以上;不定芽在培养基1/2 MS+NAA 3 mg/L+6 BA 0.1 mg/L上暗培养15 d,然后转入不加任何激素的1/2 MS培养基中,15 d即可出根,生根率为33%;生根苗栽于混合基质中,成活率可达85%。[结论]该研究建立了紫花含笑的组织培养技术。
