裂叶牵牛突变体的研究

摘    要:目的]研究裂叶牵牛花色、叶色等变化机理。方法]通过田间调查,利用PCR技术从裂叶牵牛蓝白相间突变株幼叶组织中扩增出目的片段,构建重组质粒pMD18-T/Tpn。结果]田间调查结果发现,变异株的F3代发生分离,叶色突变并不影响花色的变化;PCR扩增结果表明,以蓝白、纯蓝、大花基因组DNA在330 bp处各有1条特异带,而春光、白雪、平安红、垂蔓都没有特异带出现。结论]变异株重组质粒序列为裂叶牵牛的转座子序列片段。

Abstract:Objective]The research aimed at investigating mechanisms of the changes of Pharbitis.nil flower colors and leaf colors.Method] The recombinant plasmid pMD18-T/Tpn was constructed by amplifying the target fragments from young leaf tissues of the blue and white mutants of pharbitis nil with PCR technology and investigating in the fields.Result] The results of investigations in the fields suggested that the mutants were segregated in F3 generation,and the mutations of leaf colors did not affect the changes ...
