产脂肪酶真菌的筛选及产酶条件优化

摘    要:目的]从富含油脂的土壤中寻找高产脂肪酶的真菌,以期为扩大脂肪酶的菌源提供材料。方法]从上海市、河北省唐山、江苏省兴化、安徽省合肥地区土样中,经富集培养、平板筛选得到22株脂肪酶产生菌株,通过复筛得到1株脂肪酶活力较高真菌。根据《常见与常用真菌》对菌株进行鉴定,并研究不同条件对菌株产酶的影响。结果]从富含油脂土壤中分离得到1株产脂肪酶能力较高菌株,根据菌落群体及个体形态,查阅相关鉴定手册,初步确定该菌株为黑曲霉。菌株产酶条件研究表明:其最适产酶条件为温度30℃,初始pH 7,装液量20%,最适碳源与氮源分别为淀粉和蛋白胨。结论]为脂肪酶产生菌以及脂肪酶的工业化生产与应用奠定了基础。
