深层发酵产樟芝菌丝体和多糖培养基的研究

摘    要:目的]加快樟芝的应用和发展。方法]选取碳源及碳源浓度、氮源及氮源浓度作单因素试验,碳源、氮源、无机盐组合和生长因子作正交试验,优化深层液体发酵法生产樟芝菌丝体和多糖的培养基。结果]玉米粉作碳源时,樟芝菌丝体干重和胞外多糖含量最大,胞内多糖次之,质量分数4.00%的玉米粉作碳源最合适。以麸皮作氮源时,菌丝体干重和胞内、外多糖含量最大,硝酸铵最少,质量分数0.8%的麸皮作氮源最合适。玉米粉和麸皮是影响菌丝干重和胞内、外多糖含量的主要因素。结论]深层液体发酵法生产樟芝菌丝体和多糖的最佳培养基组合为:4.00%玉米粉,0.60%麸皮,12mg/LVB1,0.25%KH2PO4,0.05%MgSO4·7H2O。

Abstract:Objective] The aim of the research was to quicken the application and development of Antrodia Camphorata.Method] Carbon sources and their concn, nitrogen sources and their concn. were selected to make a single factor test. And carbon sources, nitrogen sources, inorganic salt combinations and growth factors were taken to make the orthogonal test. And the media for producing the mycelia and polysaccharide of A. Camphorata were optimized by submerged liquid fermentation method. Result] When corn meal was taken as carbon source, the dry weight of mycelia and the content of extracellular polysaccharide in A. Camphorata were most, followed by the intracellular polysaccharide. The optimum carbon source was corn meal with mass fraction of 4%. When wheat bran was taken as nitrogen source, the dry weight of mycelia and the content of extracellular and intracellular polysaccharides were most, the ammonium nitrate was least. The optimum nitrogen source was wheat bran with mass fraction of 0.8%. Corn meal and wheat bran were the main factors that affected the dry weight of mycelia and the contents of extracellular and intracellular polysaccharides. Conclusion] The optimum medium combination for producing the mycelia and polysaccharides in A. Camphorata by submerged liquid fermentation method were as follows:4.00% corn meal, 0.60% wheat bran, 12 mg/L VB1,0.25% KH2PO4 and 0.05% MgSO4·7H2O.
