红掌工厂化育苗关键技术研究

摘    要:目的]研究红掌工厂化育苗的关键技术。方法]以红掌盆花和切花品种的叶片、叶柄、花苞片和花苞柱头为外植体,进行组培养快速繁殖技术研究,探讨外植体的选择和消毒条件以及不同培养基对红掌愈伤组织的诱导、增殖和生根影响。结果]叶柄为最佳外植体,愈伤诱导率最高,达78.3%;外植体采用0.1%HgCl2消毒8 min的效果最好;粉冠军和MIDORI的最佳愈伤组织诱导培养基分别为1/2MS+6-BA1.50 mg/L+2,4-D0.50 mg/L+NAA0.10 mg/L和1/2 MS+6-BA1.50 mg/L+KT1.00 mg/L+2,4-D0.10 mg/L;最佳愈伤组织继代培养基分别为MS+6-BA1.00 mg/L+NAA0.50 mg/L+2,4-D0.30 mg/L和MS+6-BA0.50 mg/L+KT0.50 mg/L+NAA0.10 mg/L,添加10%的椰子汁或香蕉浆有利于增殖和丛芽生长,增殖率最高达2.83倍;最佳生根培养基为1/2 MS+NAA0.10 mg/L+IAA0.10 mg/L,生根率达100%。结论]该结果为红掌工厂化育苗体系的建立奠定了基础,对确保红掌产业的发展具有重要意义。
