摘    要:目的]建立一种检测四环素类抗生素的新方法。方法]四环素通过两步衍生化后,与氨基化的BSA上的表位氨基反应,合成TC-BSA偶联物。以四环素多克隆抗体-胶体金复合物作为分析探针,四环素完全抗原作为竞争抗原,构建分析体系。结果]紫外吸收光谱、凝胶色谱图和红外光谱均表明人工完全抗原偶联成功。选择柠檬酸三钠加入量1.5 ml生成的胶体金作为该试验继续合成胶体金检测探针的金溶胶。当抗体稀释倍数为1∶100、pH值为8.70时,金标灵敏度好。金标试纸条检测样品中四环素的检测限量是200μg/L,检测时间不超过15.0 min。与ELISA法检测样品作对比,采用新法的准确率达100%。探针溶液中加入10%(V/V)保护剂后,试剂条稳定性明显提高。结论]该研究为四环素的快速检测提供了依据。

Abstract:Objective] The study was to establish a new method for the rapid detection on tetracycline antibiotics.Method] Through two step derivatization,the tetracycline reacted with epitope amino in amination BSA to synthesize into TCBSA conjugate.With the compound of tetracycline polyclonal antibody-colloidal gold as the analysis probe and the complete antigen of tetracycline as the competitive antigen,the analysis system was constructed.Result] Ultraviolet absorption spectrum,gel chromatogram and infrared spectrum all showed that the artificial complete antigen was successfully coupled.The colloidal gold generated by adding 1.5 ml trisodium citrate was selected as the colloidal gold solution in the experiment of continuously synthesizing the colloidal gold detection probe of gold solution.When the dilution multiple of antibody was 1∶100 and pH was 8.70,the sensitivity of gold labeled strip was good The detection limit of gold-labeled reagent strip on tetracycline in detected sample was 200 μg/L and the detection time was less than 15.0 min.Compared with result of detecting samples by ELISA the accurate rate by using new method was 100%.After 10%(V/V)protective agent was added into probe solution,the stability of reagent strip was improved obviously.Conclusion] The study provided a base for the rapid detection on tetracycline.
