荷叶总黄酮的提取工艺研究

摘    要:目的]为荷叶的大规模开发利用提供技术保证。方法]采用超声波辅助萃取法对荷叶总黄酮进行提取,并用分光光度法测定其含量,通过单因素及正交试验确定最佳提取工艺。结果]当乙醇浓度达到60%时,总黄酮收率最大;液料比在10:1-30:1时,黄酮的收率随液料比的增大而增加较快,当液料比大于40:1后,黄酮收率趋于恒定;当作用时间为35min时,黄酮收率最大,但此时与30min相比,仅略微增加;提取时适宜的温度应为40℃;影响荷叶总黄酮收率的主次因素顺序为乙醇浓度〉超声时间〉液料比〉超声温度。结论]荷叶总黄酮最佳提取工艺:将荷叶干粉按35:1的液料比加入浓度60%乙醇水溶液,浸泡24h,在超声波清洗槽中超声提取35min,超声温度35℃。在该条件下,荷叶总黄酮收率为13.68%。
