摘    要:目的]研究不同激素配比对大豆子叶形成愈伤组织的影响。方法]以高蛋白大豆东农42号子叶为供试外植体,以6-BA2.0mg/L+NAA0.2 mg/L和2,4-D2.0 mg/L+KT1.0 mg/L的激素对其进行诱导,研究不同激素对大豆子叶愈伤组织的诱导效果。结果]以添加6-BA2.0 mg/L+NAA0.2 mg/L培养基培养的材料诱导率为71.4%;以添加2,4-D2.0 mg/L+KT1.0 mg/L培养基培养的材料诱导率为78.6%,诱导产生的愈伤组织大部分呈绿色、质密;未添加激素组诱导率为85.7%,但无生长迹象。所加激素均能诱导大豆子叶形成愈伤组织,但不同激素种类诱导愈伤组织能力有差异,以添加2,4-D2.0 mg/L+KT1.0 mg/L的效果最好。结论]不同种类的外源激素对外植体的诱导作用不同,大豆子叶愈伤组织诱导的最佳培养基为MS+2,4-D2.0 mg/L+KT1.0 mg/L。

Study on the Effect of Different Hormones Combinations on the Callus Induction of Soybean Cotyledon

Abstract:Objective] The effect of the different hormones on the callus formation from soybean cotyledon culturing was studied.Method] The cotyledon of the soybean variety—Dongnong No.42 with high content of protein was cultured in the medium containing two combinations of growth regulators: 2.0 mg/L of 2,4-D+1.0 mg/L of KT and 2.0 mg/L of 6-BA + 0.2 mg/L of NAA to analyze the induction efficiency of the callus formation from cotyledon.Results] In the medium with 2.0 mg/L of 6-BA + 0.2 mg/L of NAA the rate of callus induction was 71.4% and in the medium with 2.0 mg/L of 2,4-D+1.0 mg/L of KT,78.6%,which callus was with the color of green and compacter;in the medium without the addition of hormone the rate of callus induction was 85.7%,which did not appear the growing sign.The addition of growth regulator could promote the callus formation,but the different hormone had variation ability in its induction and the best induction efficiency was from the medium with 2.0 mg/L of 2,4-D+1.0 mg/L of KT.Conclusion] The induction of soybean cotyledon callus varied with the addition of hormone and the best medium for its induction was the medium MS with 2.0 mg/L of 2,4-D+1.0 mg/L of KT.
