摘    要:目的]筛选铁皮石斛离体培养开花诱导的适宜激素因子。方法]采用铁皮石斛茎段离体培养的试管苗为试验材料,以MS为基本培养基,进行单因子激素处理(PP333、TDZ的不同浓度梯度)和多因子激素处理(PP333、TDZ、6-BA以及NAA的不同组合)的比较试验,研究激素因子对铁皮石斛离体培养开花诱导的效应。结果]单因子激素处理中,PP333、TDZ的适宜浓度和其诱导的花芽形成率分别为0.2 mg/L和8.5%、0.06 mg/L和15.5%;多因子激素处理对开花诱导的效应依次为:PP333+6-BA+NAA+TDZ组合处理〉PP333+6-BA+NAA组合处理〉PP333+6-BA组合处理和PP333+NAA组合处理;适宜的激素组合为PP3330.3 mg/L+6-BA 0.5 mg/L+NAA 0.5mg/L+TDZ 0.06 mg/L,其花芽形成率和花开放率高达80.4%和90.3%。结论]PP333和TDZ对铁皮石斛离体培养开花诱导具有重要影响,TDZ的效应大于PP333。适宜的TDZ浓度与其他激素的合理配比,有利于花芽的形成。
