槟榔叶绿素含量测定影响因子研究

目的]研究槟榔叶片叶绿素含量测定方法。方法]以1年生槟榔幼苗的叶片为材料,研究不同称样量、提取时间、溶剂及光照和高温处理对其叶绿素含量测定结果的影响。结果]提取剂体积相同时,称样量越少,提取液中叶绿素含量越高;提取时间越长,叶绿素含量越高,但超过24 h后叶绿素含量开始下降;高温短时间提取比低温长时间提取叶绿素含量高,避光提取比见光提取叶绿素含量高;在相同时间和温度条件下,不同浸提剂对叶绿素的浸提效果不同,其中100%丙酮和100%乙醇以3∶1体积比混合的浸提剂提取效果最好。结论]该研究为槟榔叶片叶绿素含量测定提供了参考。
