摘    要:利用PCR-RFLP方法对3个地理居群泥蚶的核糖体DNAITS区进行分析,从分子水平探讨三者的遗传多样性及亲缘关系。结果表明:在选用的6种限制性内切酶中,BstHHI、HaeIII和Sse9I为多态性的限制性内切酶,MspI、RsaI和TaqI为单态性限制性内切酶。对酶切图谱进行数理统计后发现宁波海区泥蚶群体内核苷酸多样性指数π值为0.0017,温州海区泥蚶π值为0.0016,而莆田海区泥蚶π值为0.0011,说明宁波海区泥蚶群体内遗传多样性最大,温州海区群体内遗传多样性次之,莆田海区群体内遗传多样性最小。依据3个地理居群泥蚶间的净遗传距离Pnet值及聚类分析结果可知,莆田与温州海区泥蚶间亲缘关系最远,宁波与温州海区泥蚶间亲缘关系最近,宁波与莆田海区泥蚶间亲缘关系介于以上二者之间。

Abstract:RFLP analysis was used to investigate the genetic diversity and phylogenetic relationship of three populations of Tegillarca granosa based on the ribosomal internal transcribed spacer (ITS) region. The results showed that BstHH I, Hae III and Sse9 I were polymorphic restriction endonucleases in PCR product digestion, while Msp I, Rsa I and Taq I were monomorphic. Nucleotide diversity index (π value) of population in Ningbo was 0.001 7, and π values in Wenzhou and Fujian were 0.001 2 and 0.001 1, respectively. It was shown that the genetic diversity was the highest in Ningbo population followed by Wenzhou population and Ningbo population. According to the P_(net) value among three populations and cluster analysis results, it was concluded that the phylogenetic relationship between Fujian population and Wenzhou population was farther than that of Fujian population and Ningbo population and the geography relationship of Wenzhou population and Ningbo population was the closest.
