坛紫菜丝状体类囊体膜和光系统Ⅱ的分离鉴定

摘    要:目的]为深入研究坛紫菜的光合生理奠定基础。方法]以坛紫菜丝状体为材料,用蔗糖密度梯度超速离心法分离纯化类囊体膜和光系统Ⅱ(PsⅡ)颗粒。结果]色素带B1的吸收峰分别位于419、498、540、565和676nm,色素带B2、B3、B2.a、B2.b、B3.a.B3.b和B3.C的吸收峰分别位于419.436、485和676nm。类囊体膜B2的放氧速率和DCIP光还原活性分别为12μmol(O2)/mg(Chl)·h]和2.3aeq/mg(cm)·h];B3的分别为37μmol(O2)/mg(chl)·h]和25ueq/mg(Chl·h)];B3-b的分别为567Ixmol(O2)/mg(Cra)·h]和168ueq/nag(Chl)·h];B3-c的分别为8.1μmol(O2)/nag(Chl)·h]和23ueq/mg(Chl)·h];B2-b只有DcIP光还原活性(168ueq/mg(Cra)·h])。结论]该研究为坛紫菜丝状体类囊体膜和光系统Ⅱ的分离鉴定提供了试验依据。

Abstract:Objective] The purpose was to lay a foundation for researching the photosynthetic physiology of Porphyra haitanesis thoroughly.Method] With the conchocelis of P.haitanesis as material,the thylakoid membranes and photosystem Ⅱ(PSⅡ) particles were isolated and purified by sucrose density gradient ultracentrifugation.Result] The absorption peaks of pigment band B1 were at 419,498,540,565 and 676 nm resp.and that of B2,B3,B2-a,B2-b,B3-a,B3-b and B3-c were at 419,436,485 and 676 nm resp.The oxygen release rate and DCIP photoreduction activity of thylakoid membrane B2 were 12 μmol(O2)/mg(Chl)·h]and 2.3 ueq/mg(Chl)·h] resp.,that of B3 were 37 μmol(O2)/mg(Chl)·h] and 25 ueq/mg(Chl)·h] resp.,that of B3-b were 567 μmol(O2)/mg(Chl)·h] and 168 ueq/mg(Chl)·h] resp.,that of B3-c were 8.1 μmol(O2)/mg(Chl)·h] and 23 ueq/mg(Chl)·h] resp.and B2-b only had DCIP photoreduction activity(168 ueq/mg(Chl)·h]).Conclusion] The research provided test basis for isolating and identifying the thylakoid membranes and PSⅡ from the conchocelis of P.haitanesis.
