摘    要:目的]研究杨树eIF5A基因的功能,为林木抗性育种研究提供参考。方法]构建植物表达载体,利用农杆菌介导法转化烟草叶盘,并对转基因烟草进行分子水平鉴定。结果]杨树eIF5A基因已经整合入烟草基因组中,且转基因烟草比野生型烟草叶片抗重金属能力提高。结论]杨树eIF5A基因能促进转基因烟草的抗胁迫能力,为林木抗逆育种机理奠定了理论基础。

Abstract: Objective ] To find the gene functions of elFSA of Populus simonii x Populus rtigra and to lay the theoretical foundation for resistant breeding of forest trees. Method ] The elFSA genes were constructed in plant expression vector and transformed into tobaccos using agrobacterium-mediated procedure. Then, the transgenic tobaccos were identified by PCR. Result] The elFSA genes were integrated into the genome of tobacco and the transgenic tobaccos all improved viability under heavy metal stress when comparing with the wild type tobacco. Conclusion] The elFSA genes have a potentiality to improve viability under a biotic stresses in transgenic plants, and they lay the theoretical foundation of tree breeding for stress tolerance.
