摘    要:目的]为大白菜珍贵遗传材料的保存提供参考。方法]以受细菌污染的大白菜非整倍体组培苗和田间植株为材料,以常规消毒处理为对照,分别采用常规消毒+培养基中添加不同抗生素和农用杀菌荆预处理+常规消毒对组培苗和外植体(取自田间植株)进行处理,研究不同处理对污染的控制效果。结果]对照组组培苗培养7、14d时,污染率分别为9.1%和36.4%,培养基申添加100mg/L氨苄青霉素可有效控制污染;常规消毒外植体接种7d后污染率迭58,8%,杀菌剂预处理+常规消毒可有效控制真菌对外植体的污染,但对细菌无明显作用。结论]培养基中添加氨苄青霉素、采用合适杀菌剂对外植体进行预处理可有效控制大白菜非整倍体组织培养过程中的污染。

Preliminary Study on Contamination Control in Tissue Culture Process of Chinese Cabbage Aneuploid

Abstract:Objective] The aim was to provide the reference for conservation of rarity genetic material of Chinese cabbage.Method] With tissue culture seedling contaminated by bacterium and the plant in field of Chinese cabbage aneuploid as the materials and normal disinfection as CK, the tissue culture seedlings were treated by normal disinfection +different antibiotics added in culture medium, and the explants (from plant in field) were pretreated by agricultural fungicide+ normal disinfection.The controlling effects of different treatments on contamination were studied.Result] When cultured for 7, 14 d, the contamination rates of tissue culture seedling in CK group were 9.1% and 36.4% resp..It had effective controlling effects when 100 mg/L ampicillin was added into culture medium.The contamination rate of explants in treatment of normal disinfection was 58.8% after 7 d of inoculation.Pretreatment with bactericides+ normal disinfection could effectively control contamination of bactericides, but had no obvious effect on bacterium.Conclusion] When ampicillin was added into culture medium, explants were pretreated by optimum fungicide, the contamination in tissue culture process of Chinese cabbage aneuploid could be effectively controlled.
