小麦矮秆新基因的RAPD标记

摘    要:以矮秆易位系山农31504-1和高秆种质系山农298为材料,利用BSA法对矮秆基因进行了RAPD标记。从300个十碱基随机引物中筛选出引物S152可在矮秆DNA池中扩增出特征带。利用100株F_2单株进行连锁分析表明,引物S152扩增的RAPD标记与矮秆基因连锁,遗传距离为10.73±3.31cM。该标记可用于分子标记辅助选择,并为矮秆基因的深入研究奠定基础。

Abstract:In this work,cross between dwarf translocation line shannong 31504-1 and high germplasm line shannong 298 was carried out for constructing F_2segrigation population,and bulked segregating analysis(BSA) was used to search for random amplified polymorphic DNA(RAPD)markers linked to the new dwarf gene.300 decamer primers were screened to identify one RAPD marker(S152_(2100))linked to the new dwarf gene.Using the F2 mapping population,the dwarf gene was shown to be closely linked to the marker S152_(2100)at a genetic dis- tance of 10.73 3.31cM.This marker can be conveniently used for maker-assisted selection in wheat breeding programs for the identification and pyramiding the dwarf gene with other genes.
