摘    要:应用酸性聚丙烯酰胺凝胶电泳(APAGE)、基因组原位杂交和RFLP标记研究了异源2号和绵阳26号在醇溶蛋白和DNA水平上的差异性。APAGE分析表明,异源2号和绵阳26号至少有9条醇溶蛋白差异带,且异源2号含有小麦背景中1RS的醇溶蛋白标记位点G1d1B3。基因组原位杂交结果证实异源2号含有黑麦的1RS,而绵阳26号则不含黑麦染色质,RFLP分析结果进一步表明,位于第1同源群的短臂RFLP探针均能

Abstract:The differences on gliadin and DNA levels of Tritic um aesitivum L.cv.Yiyuan 2 and Mianyang 26 were investigated by using acid pol yacrylamide-gel electrophoresis(APAGE),genomic in situ hybridization (GISH) and RFLP markers.At least 9 different gliadin bands were identified between Yiyu an 2 and Mianyang 26.Yiyuan 2 had the gliadin marker G1d1B3 for presence of 1RS in wheat background,but Mianyang 26 did not have.Genomic in situ hybridizati on also indicated that Yiyuan 2 had the 1RS from rye and Mianyang 26 did not hav e any alien chromatin from rye.All the probes,which had been identified on the short arm of wheat homologous group 1,revealed the polymorphism between Yiyuan 2 and Mianyang 26 in RFLP analyses,derived from the missing of 1BS restriction fr agment in Yiyuan 2.These results suggested that Yiyuan 2 had the 1BL.1RS transl ocation chromosome and Mianyang 26 did not have,which was the reason of the gene tic differences between Yiyuan 2 and Mianyang 26.
