产甲烷细菌的筛选及培养技术优化研究

摘    要:采用厌氧培养技术,从贵阳青岩沼气池和花溪荷花池的渔塘污泥中分离产甲烷细菌,同时对产甲烷细菌的培养基及分离方法进行优化。经过初步的形态、生理生化特征研究,获得了3株产甲烷细菌。在培养基中同时加入营养基质甲酸钠、乙酸钠、甲醇以及注入氢和二氧化碳的混合物,使以各种生长基质为底物的产甲烷细菌在一种培养基比较容易被分离出来。用装有蒸馏水的烧杯模拟水浴锅的分离方法使分离产甲烷细菌的过程更简单、更容易操作,且整个过程不易被氧污染。并且初步筛选获得的3株产甲烷细菌在优化的培养基中生长良好。

Abstract:Three methanogenic bacteria strains were obtained from the samples collected from Qingyan biogas plants and Huaxi Hindu lotus pond and their characters of morphology,physiology and biochemistry were preliminary studied.The methanogenic bacteria strains with different growth substance were easily isolated on the media with different chemical composition.The new isolating technique had the feature of simple procedure,easy operation and without oxygen contamination.
