摘    要:应用比较基因组学和生物信息学方法,比较了从藻类到高等植物Mlo基因的CDS序列,并对该基因的遗传多样性、氨基酸功能结构及分子系统发育进行分析.结果表明,85条基因序列共检测到57个多态位点,生成70个单倍型.物种间及物种内Mlo基因存在丰富的遗传多态性;氨基酸结构域分析发现,20个物种含有一段特定的Mlo结构域,其中低等植物的跨膜螺旋区域(1~5)明显比高等植物的跨膜螺旋区域(6~8)少;分子系统进化树结果显示,不同物种的聚类情况与植物学分类系统基本一致.本研究为进一步探索Mlo基因的生物学功能和确定该基因作为分子标记提供了基础依据.

Abstract:In this study,the genetic diversity,amino acids functional structure and molecular phylogeny of the Mlo gene CDS sequences in different species,which included algae and higher plants,were analyzed using the method of comparative genomics and bioinformatics.The results showed that a total of 57 polymorphic sites were detected from 85 sequences,from which 70 hapolotypes were sorted.More nucleotide diversity and amino acid diversity of Mlo gene existed within and among species.The analysis of amino acids structural domain showed Mlo specific domain and the transmembrane helix regions of lower plants(1-5) were less than higher plants(6-8).The result of the molecular phylogenetic tree showed that the cluster situation of different species was basically consistent with botany taxonomy.This research provided the basic data for the further study on biological function of Mlo gene and the identification of molecular marker of Mlo gene.
