大岩桐离体培养技术研究

摘    要:以大岩桐带芽嫩茎为外植体,探讨不同外源激素水平对其不定芽分化、增殖与不定根形成的影响。结果表明:MS 噻二唑苯基脲(TDZ)1.0~2.0 mg/L IBA 0.1~0.3 mg/L培养基不定芽分化、增殖效果好,分化率达83.3%以上,增殖系数达6.0以上;1/2MS IAA 0.4 mg/L或1/2MS NAA 0.4 mg/L或1/2MS IBA 0.8 mg/L的生根效果好,生根率可达95.6%以上;在泥炭∶珍珠岩=1∶2的基质中驯苗,成活率可达95.3%。

Abstract:Sinningia hybrid stem were used as the explant,exploring the effects of external hormone on adventitous buds differentiation multiplication and adventitious roots forming.The results show: In the medium of MS TDZ 1.0-2.0 mg/L IBA 0.1-0.3 mg/L,the effect on differentiation and multiplicationh of sprouts is good;the differentiation rate reach above83.3 % and multiplication rate reach over 6.0;in 1/2MS IAA 0.4 mg/L or 1/2MS NAA 0.4 mg/L or 1/2MS IBA 0.8 mg/L,the effect on radication is good;the rooting rate above a level of 95.6 %;using stroma of perlitehumus soil is 1:2 raising plants.the survival rate can reach a level of 95.3 %.
