兔胚胎玻璃化冷冻的研究

摘    要:探讨用玻璃化冷冻法保存兔胚胎的效果.结果表明,兔早期囊胚在0.5M海藻糖(93.8%)溶液中的存活率显著高于0.5M蔗糖组(70.0%,P〈0.05).对照组与不同浓度甘油处理间兔胚胎存活率均差异不显著(P〉0.05).分别用不同浓度乙二醇玻璃化冷冻兔胚胎,解冻回收后正常胚胎数和囊胚孵化率,对照组与10%,20%EG组之间均差异不显著(P〉0.05),但对照组,10%,20%EG组均显著高于30%EG组(P〈0.05).解冻回收后正常胚胎数体内胚胎组(79.4%)显著高于体外胚胎(54.1%,P〈0.05),囊胚孵化率2组之间差异不显著(P〉0.05).缓慢冷冻法与玻璃化冷冻法之间胚胎解冻回收后正常胚胎数和囊胚孵化率均差异不显著(P〉0.05).

Abstract:Explore the preservation of rabbit embryos effects adopting vitrification freezing. The results showed that the survival rate of rabbit early blastocysts treated with 0.5 mol/L trehalose was significantly higher than that treated with 0.5 mol/L sucrose (93.8% vs. 70.0%, P〈0.05). There were not significantly different between control group and other treatments of different concentration of glycerol on the survival rates of rabbit early blastocysts (P〉0.05). Frozen with different concentration of EG, there is no significantly different between the control group and 10% EG, 20% EG after thawing on the normal embryo rate and hatching rate of blastula (P〉0.05), but both the normal embryo rate and hatching rate of blastula of all the three groups were significantly higher than 30% EG group after thawing (P〈0.05). In the experiment of the frozen effects on rabbit embryos in vivo and in vitro, the normal embryos rate was 79.4% and 54.1%, respectively after thawing, and the former was significantly higher than the latter (P〈0.05). There was not significant difference on the hatching rate of blastula between the two groups (P〉0.05). In the experiment of researching frozen effects on rabbit embryos treated with different freezing methods, there was not significant difference on the normal embryo rate and hatching rate of blastula between slow freezing and vitrification freezing after thawing (P〉 0.05).
