| 摘 要： | 采集多浪羊的外周静脉血液,用人淋巴细胞分离液进行分离,收集分离得到淋巴细胞,同时经过纯化后通过镜检观察细胞形态。将纯化后的淋巴细胞在RPM I1640中置37℃,5%CO2培养箱4,6,8和12 h进行培养并观察淋巴细胞,其存活率分别为93.1%、92.4%、91.5%和90.6%。本试验成功地建立了多浪羊外周血淋巴细胞的分离技术和体外短期培养体系。

| Abstract: | Ovine peripheral blood were collected from Duo Lang sheep and separated by using lymphocytes separation medium.Isolated lymphocytes were purified.Then the cell was observed by microscope.Purified lymphocytes were cultured in RPMI1640 at 25 ℃,5% CO2 for 4,6,8 and 12 h and the survival situation of lymphocytes was observed.The survival rate of leucocytes were separately 93.1% 、92.4%、91.5% and 90.6%.By the experiment,the methods of leucocytes separation from ovine peripheral blood and an efficient method to culture ovine peripheral blood leucocytes in vitro were successfully established. |
