摘    要:本试验采用三种不同的方法提取葡萄幼叶基因组DNA,即:改良的CTAB法、高盐法和SDS法。并通过紫外/可见分光光度计检测、琼脂糖凝胶电泳检测、RAPD-PCR扩增对三种方法提取的DNA进行定量、定性分析和综合比较。结果表明,三种不同的方法提取葡萄幼叶DNA均能满足RAPD扩增要求,扩增效果为:CTAB高盐法SDS法。因此,改良的CTAB法是一种适合新疆葡萄基因组DNA提取的最佳方法。

Abstract:Total DNA from the young leaves of grape was extracted using three different methods, including the CTAB, high salt and SDS methods. The resulted DNA samples of three methods were tested qualitatively and quantitatively using the value of optical density (OD) , agarose gel electrophoresis and RAPD. The results showed that there were three different methods to meet the requirement of RAPD amplification. The amplification effect of extracted DNA was CTAB 〉 high salt 〉 SDS method. So, method of improved CTAB was the best method to extract genomic DNA of the Xinjiang grape.
