针刺联合亚低温对脑缺血/再灌注损伤大鼠Bcl-2、Bax、Caspase-3蛋白表达的影响

摘    要:目的 探讨针刺联合亚低温方法对脑缺血/再灌注(ischemia/reperfusion,I/R)损伤大鼠梗死面积比及Bcl-2、Bax、Caspase-3蛋白表达的影响。方法 将50只SD健康雄性大鼠常规饲养1周后,随机选取假手术组10只,余40只用Zea Longa线栓法制作大鼠大脑中动脉闭塞局灶脑I/R模型,待造模成功后,再将40只SD大鼠随机分为模型组、针刺组、亚低温组、针刺联合亚低温组,每组各10只。治疗72 h后,使用TTC染色检测脑梗死面积比、免疫组化法检测Bcl-2、Bax、Caspase-3蛋白的阳性细胞数。结果 与假手术组比较,模型组大鼠梗死面积比、Bax、Caspase-3蛋白表达水平明显增高,Bcl-2表达水平显著下降,差异有统计学意义(P<0.05或P<0.01);与模型组比较,各治疗组大鼠梗死面积比、Bax、Caspase-3蛋白表达水平明显降低,Bcl-2表达水平明显升高,差异有统计学意义(P<0.05或P<0.01);各治疗组之间Bcl-2、Bax、Caspase-3的差异无统计学意义(P>0.05),但脑梗死面积比针刺联合亚低温组较针刺组和亚低温组低,差异有统计学意义(P<0.05或P<0.01)。结论 针刺联合亚低温治疗可通过减少脑梗死面积比、提高Bcl-2表达水平、降低Bax与Caspase-3蛋白表达从而实现对脑细胞的保护作用。

Effect of Acupuncture combined Hypothermia on Bcl-2, Bax and Caspase-3 Expressions of Cerebral Ischemia Reperfusion Injury Rats

Abstract:Objective To investigate acupuncture combined hypothermia on infarction area ratio, Bcl-2, Bax and Caspase-3 protein expressions in rats with cerebral ischemia/reperfusion injury. Methods 50 healthy male SD rats were given conventinal feeding for 1 week, then 10 rats were randomly selected as the sham-operation group, the other 40 rats were built middle cerebral artery occlusion focal cerebral ischemia/reperfusion models by Zeab Longa suture method. After the success of the modeling, then 40 SD rats were randomly divided into model group, acupuncture group, hypothermia group, the acupuncture combined hypothermia group, 10 rats in each group. After treatment for 72 h, the cerebral infarction area ratio was detected by TTC staining method. The apoptosis cell numbers of Bcl-2, Bax and Caspase-3 proteins were detected by immunohistochemical method. Results Compared with the sham-operation group, the infarction area ratio, Bax, Caspase-3 protein expressions in rats of model group were increased obviously, the Bcl-2 expression level decreased significantly, the difference was statistically significant (P<0.05 or P<0.01); Compared with the model group, the infarction area ratio, Bax, Caspase-3 protein expressions in each treatment group were decreased obviously, the Bcl-2 expression level decreased significantly, the difference was statistically significant (P<0.05 or P<0.01); The indexes of Bcl-2, Bax and Caspase-3 in each treatment group have no statistical significance (P>0.05), but the trend was that acupuncture combined hypothermia group was superior to the acupuncture group and hypothermia group. Conclusion Acupuncture combined hypothermia treatment can reduce the cerebral infarction area ratio, improve the level of the Bcl-2 expressions and reduction of Bax and Caspase 3 protein expressions to protect brain cells.
