甘蓝型黄籽油菜原生质体的游离和纯化

摘    要:以西南农业大学选育的渝黄1号和SH212个甘蓝型黄籽油菜品种为材料,切取其无菌苗的子叶和下胚轴分离纯化原生质体。种子萌发时低温处理可明显提高原生质体活力;光照处理对子叶原生质体活力有较大影响;酶液配比对原生质体产率影响较大;用“过滤-离心-漂浮法”进行纯化,用21%蔗糖,800转/min纯化效果较好。

The proplast isolation and purification of yellow-seed Rape

Abstract:Proplasts were isolated and purified from hypocotyls and plantlet of two yellow-seed (Brassica napus L.) cultivars SH21 and Yuhuang No.1 .Low temperature treatment during seed generation could apparently increase the vigour of protoplasts. The yield of protoplasts was effect by different enzyme. The method of purification called filtrate-centrifugl-float, floated by 21%sucrose and centrifugalized at 800 r/min is better.
