ACC脱氨酶活性菌株的筛选、鉴定及其对茄子耐盐性的影响

摘    要:    从浙江上虞海涂土样中分离、筛选到1株具有1-氨基环丙烷1-羧酸(ACC)脱氨酶活性的菌株DW1根据生理生化特征、16S rDNA序列分析、(G+C)mol%测定,菌株DW1被鉴定为假单胞茵(Pseudomonas spDW1)研究该菌株对茄子耐盐性的影响,结果表明:用茵株DW1处理可以提高种子的发芽势及芽长;在茄苗盆栽实验中,经DW1菌液处理的种子,在高盐浓度下茄苗的鲜重与对照相比有显著提高,同时Ca2+含量有所增加而Na+含量则有所下降,其茄苗的长势明显优于对照说明用具有ACC脱氨酶活性的菌株DW1处理可以有效地提高茄苗的耐盐性

Abstract:A bacterium, named strain DW1 with 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity, was isolated from saline soil in Shangyu, Zhejiang Province. Based on the results of phenotypic features, phylogenetic similarity of 16S rDNA gene sequences, DNA G C content, strain DW1 was identified as Pseudomonas sp. The response of eggplant seeds and seedlings treated with strain DW1 to salt stress was assessed. The results show that strain DW1 was capable of promoting eggplant seed germination percentage and seedling root elongation compared with control. Interestingly, the strain DW1 significantly increased the fresh weights of eggplant seedlings in saline soil and slightly influenced the uptake of calcium ions while reduced the uptake of sodium ions. This may suggest strain DW1 can increase resistance in eggplant to salt stress.
