石刁柏胚状体诱导与影响因素的研究

本试验对石刁柏体细胞胚及花粉胚状体诱导进行了研究。试验表明，不同基因型对胚状体发生频率影响很大，接种材料不同会导致胚性愈伤组织形成上的差异，外源物质及植物激素种类、配比对胚状体诱导、分化起着重要作用。ＭＳ培养基附加６％蔗糖，水解酪蛋白６００ｍｇ／Ｌ，谷氨酰胺７００ｍｇ／Ｌ，ＩＢＡ２．５ｍｇ／Ｌ，ＢＡ０．８ｍｇ／Ｌ，胚性愈伤组织诱导率最高；而ＭＳ＋ＮＡＡ０．３ｍｇ／Ｌ＋ＫＴ０．２ｍｇ／Ｌ＋嘧啶醇０．８ｍｇ／Ｌ培养基，胚状体诱导效果最佳，平均每块愈伤组织形成体细胞胚１．７２个，花粉胚状体１．４６个。

Studies on Embryoid Induction of Asparagus officinalis L.and the Influencing Factors

The induction of body cell embryo and anther embryoid of Asparagus officinalis L. was studied.Results were the following:The embryoid forming frequency varied considerablly in different genotypes.Various materials led to diversity in embry-onic calli formation.The contents of external substances and plant hormones had im-portant impacts on embryoid induction and differentiation. The MS medium, addingsugar 6%, casein hydrolystate 600mg/L,Gln 700mg/L, IBA 2.5mg/L, BA0.8mg/L, induced embryonic calli in the highst rate. Emhryoid induction required rela-tively lower concentrations of plant hormones, the optimum effect occurred in theMS medium +NAA 0.3mg/ L+KT 0.2mg/L+Pyrimidine 0.8mg/L. On every calliumthere formed 1.72 body cell embryoes and 1.46 anther embryoids.
