刺五加叶片组织培养研究

摘    要:为扩大刺五加种群数量,以刺五加叶片、叶柄为外植体,以MS为基本培养基,研究了不同调节因子组合对愈伤组织诱导、分化的影响。结果表明,诱导刺五加叶片、叶柄形成愈伤组织的最佳培养基为MS+1.5 mgL-16-BA+1.5mgL-12,4-D,诱导率分别为86.70%、46.70%。最佳分化培养基为MS+2.0 mgL-16-BA+0.20 mgL-1NAA,叶片分化率达80.80%,叶柄分化率为53.33%,愈伤块形成芽丛的情况最好。

Abstract:The major object of this study is to expand population of Acanthopanax senticosus.We used its leaf and petiole as explants for the first time In this paper,we adopted MS as basic medium and studied induction and differentiation of cullus by different combination of,controlled factors.The result showed that the optimal medium for the leaf,petiole of Acanthopanax senticosus was MS 1.5mgL-16-BA 1.5mgL-1 2,4-D,and the inducement rate of bublets was 86.70%,46.70% respectively.The preferable medium of differentiation was MS 2.0mgL-16-BA 0.20mgL-1NAA,and the bud differentiation rates of leaf and petiole were 80.80%,53.33% respectively.
