猪肺炎支原体、鸡毒支原体膜蛋白免疫原性分析

摘    要:通过分析猪肺炎支原体(Mhp)、鸡毒支原体(MG)膜蛋白的免疫原性及化学成分,为其致病机理的研究提供基础。分别采用SDS-PAGE、免疫印迹、高碘酸雪夫染色(PAS法)和高碘酸一硝酸银法对Mhp232株和MGFMF4株膜蛋白的分子量范围、种类和免疫原性进行分析,鉴定膜糖蛋白的数量与种类。Mhp膜蛋白分子量在30~250ku之间,有12条带;MG膜蛋白分子量在25~200ku之间,有29条带。免疫印迹表明,Mhp中46ku膜蛋白具有免疫原性;MG中56ku膜蛋白具有免疫原性。PAGE电泳表明,Mhp有5条带,MG有12条带。PAS法鉴定膜糖蛋白,Mhp有1条带,MG有2条带;高碘酸一硝酸银法鉴定膜糖蛋白,Mhp约在相同位置出现1条带,而MG显示7条带,表明此法的灵敏度高于PAS法。

Immunogenicity Analysis on Membrane Proteins of Mycoplasma hyopneumonia and Mycoplasma gallisepticum

Abstract:The pathogenic mechanism research of Mycoplasma hyopneurnoniae (Mhp) and Mycoplasma gallisepticurn (MG) were conducted by analyzing the immunogenicity and chemical composition of the membrane proteins of Mhp and MG with SDS-PAGE, Western-blot, PAS and periodic acid-silver nitrate method. The molecular weight range of the Mhp 232 strain membrane proteins was between 30-250 ku and there were twelve bands. The molecular weight range of MG FMF4 strain was between 25-200 ku, and there were twenty nine bands. The test results with Western-blot a- nalysis indicated that the 46 ku membrane protein in Mhp and the 56 ku membrane protein in MG had immunogenicity. Mhp showed five bands, and MG showed twelve bands with PAGE electrophoresis. The membrane glycoproteins were identified using PAS method in which Mhp had one band and MG two bands. When using the periodic acid-silver nitrate method, Mhp showed one band almost at the same position in gel as with PAS method, and the MG had seven bands which indicated that this method was more sensitive than PAS method.
