大豆外植体培养再生植株的研究

摘    要:本文报道大豆小真叶和上胚轴外植体培养成苗的途径,并讨论了外源激素对苗分化的效应。大豆小真叶外植体在合有NAA或KT+NAA的B_5培养基上,上胚轴外植体在含有KT+NAA的B_5培养基上,可脱分化形成愈伤组织,并由愈伤组织直接分化成苗。小真叶外植体在含有KT+NAA的B_5培养基上比在只含有NAA的B_5培养基上成苗率高。在KT+NAA的B_5培养基上,上胚轴较小真叶外植体成苗率高。 NAA对促进小真叶外植体分化根较诱导愈伤组织和芽的分化更有利。KT对诱导愈伤组织和芽的分化有利。KT与NAA配合使用对大豆外植体培养再生植株较单独使用效果好。在配合比例中,NAA浓度较低的比NAA浓度较高的芽分化早,成苗率高,后者分化的根较多。


A STUDY OF THE REGENERATION OF PLANTLETS FROM SOYBEAN EXPLANT CULTURES
Abstract:The purpose of this paper is to introduce the ways of how the young ture leaves and the epicotyls of soybeans are cultured into plantlets and discuss the effect of plant growth hormones on shoot differentiation from explant. The young true leaves explant of soybean are cultured in B_5 basal medium containing NAA or KT+NAA,epicotyls explants are cultured in the B_5 medium containing KT+NAA, they are able to dedifferentiate into callus tissues, then directly develop into plantlets. If young true leaves are cultured in the B_5 medium containingKT+NAA, the rate of development of plantlets is higher than that only containing NAA. However, in the B_5 medium which contains KT+NAA, the rate of plantlets developed from epicotyl is higher than that from true leaves. It is more effective that NAA can accelerate the differentiation roots from young true leaves than it inducts callus tissues and the differentiation of plantlets, but KT is beneficial to the induction of callus tissues and the differentiation of plantlets. In the regeneration of plantlets from soybean explant, the result in the KT+NAA is better than only in the KT. In the compound ratio of KT+NAA, plantlets are differentiated earlier in lower concentration of NAA than in higher concentration, and the rate is higher too, however, more roots are differentiated in the later.
