黄花柳中总黄酮含量的研究

采用乙醇回流提取法,以亚硝酸钠、硝酸铝、氢氧化钠为试剂,芦丁为对照研究黄花柳花穗中黄酮类化合物的最佳提取工艺条件。用正交实验法对黄花柳花穗中的黄酮类化合物的提取工艺进行了优化研究,采用正交表L9(34),以乙醇浓度、回流温度、回流时间及料液比为因素,以总黄酮显色液的吸光度为指标进行实验。得到的黄花柳花穗中总黄酮的最佳提取工艺条件为:乙醇浓度为50%,料液比为1∶40,回流温度为70℃,回流时间为3 h,在最佳工艺条件下的黄酮含量为26.82 mg/g。实验方法简单、方便,结果准确、可靠,最佳条件适合批量生产中该药材的提取。

Study on Total Flavonoid Content from Salix Caprea

In this paper,the extraction of flavonoids from Salix caprea flowers was studied,with ethanol as solvent,sodium nitrite,aluminium nitrate and sodium hydroxide as indicator,rutin as control sample.The optimum technology of extracting flavonoids in Salix caprea flowers was studied by orthogonal test.An orthogonal test table L9(34) was used with the factors of concentration of extracting solution,temperature,time and solid-liquid ratio.The content of flavonoids was determined by spectrophotometry.The optimum technology is as follows: the methanol concentration is 50%,the ratio of mass of raw material to the volume of solvent(solid-liquid ratio) is 1∶40,refluxing time is 3 h and refluxing temperature is 70℃.According to the conditions mentioned above,flavonoids were extracted efficiently and the extraction yield of the flavonoids could reach 26.82 mg/mL.The method is simple,convenient,and can be applied to extract flavonids in large scale.
