微卫星标记BM6506和BMS1788的多态性与新疆山羊绒性状的关系

摘    要:目的]研究微卫星标记BM6506和BMS1788在新疆山羊群体中的遗传多态性与经济性状的关联性,以期找到与生产性状相关的遗传标记,为标记辅助选择育种提供理论依据.方法]采用微卫星标记技术研究微卫星标记BM6506和BMS1788在新疆山羊群体中的遗传多态性,利用POPGENE(Versionl.32)软件计算等位基因频率(E)、基因杂合度(He)、有效等位基因数(Ne)、基因纯和度(P);用PIC-CALC软件计算多态信息含量(PIC),运用最小二乘线性模型,对所检测到的多态标记与生产性状进行关联分析.结果]2个微卫星标记在新疆山羊群体中的有效等位基因数(Ne)分别为4.162 5、5.732 5,基因杂合度(He)分别为0.759 8、0.825 6,多态信息含量分别(PIC)为0.720 8、0.801 6.关联分析结果表明-在绒细度性状上:标记BMS1788的CF和EG基因型个体显著高于AE基因型个体(P<0.05);在绒长度性状上:标记BM6506的AB基因型个体显著高于BB、BD和AA基因型个体(P<0.05),标记BMS1788的CF基因型个体显著高于AD和DE基因型个体(P<0.05);在含绒率性状上:标记BM6506的AC基因型个体显著高于BB和AA基因型个体(P<0.05).结论]微卫星标记在BM6506和BMS1788在新疆山羊群体中均表现为高度多态(PIC>0.5),表明新疆山羊群体的遗传变异程度大,遗传多样性丰富,可以用作新疆山羊遗传多样性的评估和经济性状选择的遗传标记.

Abstract:【Objective】The research aims to study the genetic polymorphism about two microsatellite markers BM 6506 and BM S1788 and analyze the correlationships between the economic traits in Xinjiang goat population in order to find out genetic marker associated with economic traits and provide certain theoretical basis for marker assisted breeding.【Method】The genetic diversity of two microsatellite markers in Xinjiang goat populations is studied by using microsatellite marker technology.Gene frequency(P),gene heterozygosity(H),effective number of alleles(Ne) are calculated by Popgene software(Version 1.32 ), and polymorphism information content(PIC) is calculated by PIC - CALC software.The correlation between microsatellite markers and economic traits is analyzed by using of least square liner model.【Result】The results showed that,in Xinjiang goat population,an effective number of alleles(Ne) is 4.162 5,5.732 5, respectively;gene heterozygosity(He) is 0.759 8,0.825 6,respectively;polymorphism information content (PIC) is 0.7208,0.8016,respectively.Correlation analysis results indicate that in fineness traits,the genotypes CF and EG of BMS 1788 was higher than that of AE genotype(P < 0.05);In cashmere length traits,the genotype AB of BM 6506 was higher than those of BB,BD and AA genotypes(P < 0.05); genotype CF of BMS 1788 was higher than those of AD and DE genotypes(P <0.05);In rate of cashmere traits,genotype AC of BM 6506 was higher than those of A A and BB genotypes(P <0.05).【Conclusion】This shows that microsatellite markers BM 506 and BMS 1788 belong to high polymorphism in the Xinjiang goat population,and they can be used as an assessment of goat genetic diversity and could be regarded as genetic marker of economic traits choice.
