向日葵黑茎病菌分离鉴定及其RFLP分析

摘    要:目的]对新疆近期发生的向日葵黑茎病进行病原菌分离鉴定,并对该菌进行RFLP分析.方法]用常规分离培养法从田间采集的向日葵黑茎病病株获得致病菌,观察该菌的形态学特征;用PCR法对该菌115区进行扩增和测序,并与向日葵茎点霉黑茎病菌进行分子比对,明确二者的同源性;选取10种限制性内切酶对该菌的ITS区PCR产物进行RFLP分析.结果]该菌的形态学特征为茎点霉属真菌,与向日葵茎点霉黑茎病菌的同源性达到99;以上,说明该致病菌为向日葵茎点霉黑茎病菌Phoma helianthi Taberosi.通过RFLP分析确定了该病菌的酶切位点.结论]该致病菌的形态学和分子生物学鉴定结果表明,向日葵黑茎病由向日葵茎点霉黑茎病菌引起.

Abstract:【Objective】The pathogen was isolated and identified which has caused sunflower black stem in Xinjiang recently.【Method】The pathogen was separated and culturated from the sunflower black stem's plants which were collected in the fields by the traditional method.The fungus' morphology characteristics were observed,its district was analysed and amplified by PCR.The pathogen's RFLP was analyzed by 10 kinds of different enzymes.【Result】The morphological observation showed the pathogen belongs to Phoma.The homology of the pathogen and Phoma helianthi Taberosi reached 99%.The results identified that it belongs to Phoma helianthi Taberosi.【Conclusion】The results indicated that the pathogen of sunflower black stem belongs to Phoma helianthi Taberosi.
