孜然芹子叶愈伤组织诱导及继代培养研究

摘    要:探讨孜然芹种子表面消毒的适宜方法,以孜然芹无菌苗子叶为外植体,研究最适宜愈伤组织诱导及继代培养的激素种类及浓度。结果表明:孜然芹种子经7 0%酒精1 0 s+0.1%升汞5 0 s,2 0 d萌发率可达97.70%,污染率11.11%;子叶切段在光照时间16 h/d,MS+2,4-D 2 mg/L的愈伤组织诱导启动培养基中,30d愈伤诱导率达100%,继代培养在MS+NAA 0.5 mg/L+6-BA 0.2 mg/L培养基中光照16 h/d培养20 d,愈伤组织增殖率达67%,愈伤组织在同种继代培养基中暗培养20 d,增殖率达80%。

Abstract:The ways of the sterilization from the seeds of Cuminum cyminum L.were researched,the callus were induced from Cotyledon culture in Vitro of Cuminum cyminum L.,the secondary culture from the callus were studied.Germination percentage of the seeds was 97.78%,and contamination percentage was 11.11% after being sterilized in 70% alcohol for 10 s,and in 0.1% HgCl2 for 50 s.The rate of the callus induction of Cotyledon culture was 100% following a period of 30 d with 16 hour photoperiod everday on MS basal medium supplemented with 2,4-D 2 mg/L.The proliferation rate of callus induction was 67%.The secondary culture was illuminated for 16 h/d on MS basal culture medium supplemented with NAA 0.5 mg/L 6-BA 0.2 mg/L to be cultured 20 d.The rate of growth of callus could be up to 80% after being cultured in a dark condition for 20 d with the same secondary culture medium.
