摘    要:【目的】探讨新疆野生石竹的系统发育和种间亲缘关系。【方法】采用CTAB+硅珠法提取总DNA,并对nrDNA的ITS区(包括ITS-1,5.8SrDNA和ITS-2)进行了序列测定。采用邻位相接法(NJ)和最大简约法(MP)进行聚类分析。【结果】新疆石竹属植物的ITS序列总长度为610~614bp,ITS-1和ITS-2序列长度为235~238bp和214~215bp。5.8S序列很保守,长度均为161bp。在整个ITS序列中,共有119个变异位点,17个信息位点,分别占整个ITS序列长度的19.71%和2.77%。【结论】齿瓣组和纟遂瓣组分别聚成一支构成姊妹群,其支持率分别为90%和98%。并对种间亲缘关系进行了分析。

Abstract:Objective and Method]The objective of this paper was to discuss the relationships between phylogensis and interspecies of wild Dianthus from Xinjiang.The total DNA were extracted with CTAB method and ITS of nrDNA including ITS-1,5.85 rDNA and ITS-2 were determined with sequences.The cluster analysis were carried out by use of maximum parsimony (MP) and Neighbour-joining(NJ).Result]The total length of ITS sequences of Xinjiang Dianthus was from 610 to 614 bp,with ITS-1 (235-238 bp),slightly shorter than ITS-2 (214-215 bp),with 119 variation sites and 17 information sites,occupied 19.71% and 2.77% of ITS.The 5.8S sequence is too conservative with all 161 bp.Conclusion]The analysis indicates that sect.Barbulatum and sect.Fimbriatum could be divided into two main clades with bootstap values of 98% and 85%.An analysis was carried out on the relationship between phylogenesis and interspecies.
