新疆野生冰草抗旱功能基因BADH的克隆及表达载体的构建

摘    要:【目的】克隆新疆野生冰草抗旱相关基因甜菜碱醛脱氢酶(betaine aldehyde dehydrogenase,BADH)基因,为进一步研究其抗旱功能奠定基础。【方法】利用同源克隆法和RACE方法扩增获得新疆草原区强抗旱植物沙生冰草抗旱相关基因BADH,并将其构建到植物表达载体pBI121上。【结果】克隆获得的新疆野生沙生冰草抗旱相关基因BADH全长1422 bp,编码区序列全长为1182 bp,GenBank注册号为GU181396.1,并成功构建了植物过表达载体。

Cloning and Plant Expression Vector Construction of BADH from Wheatgrass in Xinjiang

Abstract:Objective]For further study of the drought resistance ability of BADH,we cloned the drought resistance related gene BADH from wheatgrass in Xinjiang.Method]Homologous cloning method and RACE amplification method were applied to clone the drought resistance related gene BADH from wheatgrass in Xinjiang.The plant expression vector of BADH was constructed and named pBI121 - BADH.Result] The sequence of BADH obtained from wheatgrass in Xinjiang clone was 1 422 bp,the ORF was 1 182 bp,GenBank registration number is GU181396.1,and the plant expression vector of BADH was constructed successfully.
