石河子地区棉田棉铃虫种群体内相关酶活及含量的测定

摘    要:【目的】测定石河子地区棉铃虫体内羧酸酯酶、磷酸酯酶、谷胱甘肽-S-转移酶以及多功能氧化酶(MFOs)四类主要解毒代谢酶的含量及活性,并与敏感品系棉铃虫进行比较,为棉铃虫抗药性研究提供实验依据。【方法】利用紫外分光光度计比色法测定酶源蛋白含量及比活力。【结果】石河子地区棉铃虫体内的MFOs的酶原蛋白含量及比活力均高于敏感品系棉铃虫且差异显著,其酶源蛋白含量分别为105.943 167、96.158 47μg/头,比活力(0.485 3±0.002 1)、(0.352 3±0.001 4)μM/μg·30 min,其余三种酶在两品系间差异不显著。【结论】NFOs活性的增加是石河子地区棉铃虫抗药性增强重要因素之一。

Determination of Contents and Activities of Detoxification Enzymes of Helicoverpa armigera in Field Populations in Shihezi District

Abstract:Objective]In order to provide experimental data for the research on the insecticide resistance of H.armigera,the contents and activities of the main detoxification enzymes such as carboxylesterase(CarE ),phosphatase,glutathione S - transferase(GST ),monooxygenase(MFOs) of H. armigera in field populations in Shihezi district were determined when compared with the susceptible strain of H.armigera.Method]The enzyme protein contents and specific activities were measured by ultraviolet spectrophotometry.Result]The content of MFOs of H.armigera in field populations in Shihezi district was 105.943 167 jxg/insect,distinctly higher than that of susceptible strain which was 96.158 47 jjug/insect. Furthermore,the specific activity of the former was 0.485 3±0.002 1μM/μg ? 30 min,also higher than that of susceptible strain which was 0.352 3±0.001 4μM/μg ? 30 min.There were no significant differences of the contents and activities of the other diree enzymes between field populations in Shihezi district and the susceptible strain.Conclusion]The increased activity of MFOs was one of the main factors contributing to the enhanced resistance to insecticides of H.armigera in field populations in Shihezi district.
