摘    要:【目的】依据形态学、生理生化和26S rDNA D1/D2区序列特征,确定一株胡杨内生酵母菌的分类学地位。【方法】通过平板划线分离法分离纯化胡杨树干部内存液样品中的内生酵母菌;检测分离菌株形态学特征和生理生化特征,进一步对酵母菌株的26S rDNA D1/D2区序列进行扩增以及系统进化分析。【结果】传统的形态学特征和生理生化特征鉴定及26S rDNA D1/D2区序列分析结果表明菌株ZA-7与Rhodotorulaminuta(FR870029)相近,相似率为99.4%,存在4个碱基的差异。【结论】从胡杨树干液中分离得到了一株尚未从植物中分离到的内生酵母菌,并将该菌株鉴定为Rhodotorula minuta的近似种。

Abstract:【Objective】 The taxonomic position of an endophytic yeast from the Populus euphratica was determined based on morphological,phenotypic and genetic characters.【Method】 Endophytic yeast strain was isolated from the storage liquid in the stem of Populus euphratica stands by using streak plate method.Its morphological,physiological and biochemical characteristics were detected,followed by an analysis of 26S rDNA D1/D2 region sequences.【Result】 The traditional morphological identification method and the sequence analysis of 26S rDNA D1/D2 showed that strain ZA-7 was closely related to Rhodotorula minuta(FR870029) with the similarity of as high as 99.4%.【Conclusion】 For the first time an endophytic yeast strain was isolated from the storage liquid in the stem of Populus euphratica stands and identified to be the most similar species of Rhodotorula minuta.
