摘    要:利用小麦穗离体培养技术和SDS-PAGE技术,研究了不同氮素浓度处理下小麦籽粒高分子量谷蛋白亚基(HMW-GS)的表达和积累规律。研究显示:氮素浓度不影响HMW-GS组成和表达顺序,各处理下HMW-GS的表达顺序均依次为亚基8、亚基7、亚基2和亚基12。但各亚基的表达时间与含量明显受氮素浓度影响:低氮浓度处理与高氮浓度处理相比,亚基8、亚基7、亚基2和亚基12的表达时间分别从花后121、5和18 d提前到花后6和9 d;HMW-GS的含量均与氮素浓度呈正相关,特别是亚基2不仅与氮素浓度呈极显著正相关,且影响亚基12对氮素的响应。

Abstract:Development and accumulation law of HMW-GS was studied in different nitrogen content by SDS-PAGE technology in wheat in vitro.The results showed that nitrogen concentration didn't affect the composition and the expression order of HMW-GS.The expression order of HMW-GS was subunit 8,subunit 7,subunit 2 and subunit 12 in turn in different nitrogen concentration,but the expression time and content of each subunit were affected by nitrogen concentration obviously.The expression time of subunit 8,subunit 7,subunit 2 and subunit 12 was advanced ahead of time after flower 6 d and 9 d from the time after flower 12 d,15 d,18 d individually in higher nitrogen content.A significant positive correlation was found between the content of HMW-GS and the different nitrogen concentration,especially the subunit 2.The subunit 2 was not only extremely remarkable positive correlation with the nitrogen concentration,but also affect the nitrogen response of the subunit 12.
