陆地棉矮化突变体及其SSR指纹分析

摘    要:利用离子束注入陆地棉种子,经过连续6~7代繁殖和筛选,发现的突变株系形态表现一致、株高≤75cm,植株后期不徒长,不发侧枝,自封顶,该突变株系命名为"矮化(免打顶)突变体"。以该棉花矮化突变体为材料,研究了SSR-PCR反应体系中各主要成分对检测结果的影响,优化了适宜棉花SSR反应体系,并从分子标记的角度表明了矮化突变体与野生型相比较,具有较多基因突变和遗传差异;从形态学的角度研究表明突变体能够自然控制植株高度,能够实现棉花突变体免打顶,减少化学试剂的用量和污染,降低农民劳动强度、生产成本,提高棉花产量、纤维品质,推动棉花生产的全面机械化和现代化有重要的现实意义。

Abstract:After ion beam being injected into upland cotton seed,the cotton was breeded 6-7 generations. Mutants were screened and named dwarfing(free topping) mutants.The impact of different major components of SSR -PCR reaction system on the test result were investigated.The molecular marker analysis indicated that the mutant possessed lots mutant genes and genic differences compared with that of the wild type.The morphological study showed the mutant plant height was controlled by themselves,that is,these cotton were free topping.It brought to reduce the amount of the chemical reagents,reduce the pollution of the environment and the amount of labor.It was important that these dwarfing cottons promoted the mechanization and modernization of cotton production.
