摘    要:本研究以红丝青叶为例,对其组培过程中出现的典型污染菌进行分离与鉴定,并通过系统进化树明确了各菌种的进化关系。结果表明,在红丝青叶组培过程中出现的污染菌种类繁多,包含细菌和真菌,细菌归类于芽孢杆菌属、肠杆菌属、泛菌属、科萨克氏菌属、剑菌属和克雷伯菌属,真菌归类于寡营养丛孢霉属、枝孢霉属和隔孢伏革菌属。本研究结果可为后期组培培养基的优化及污染菌的防治提供参考。

Preliminary isolation and identification of typical bacteria contaminated by tissue culture of aquatic#$NBSplant
Abstract:In this study, the contamination of Hygrophila polysperma was investigated, and the results showed that various species of bacteria and fungi were identified in culture medium. The bacterial or fungal strains were isolated from LB or PDA medium and identified on the observation of colonial morphology and further analyzed based on the homology of 16S rDNA or ITS sequences. Among them, bacteria belong to six genera (Bacillus, Enterobacter, Pantoea, Kosakonia, Ensifer and Klebsiella); fungi were divided into three genera (Plectosphaerella, Cladosporium and Peniophora). This study would lay the foundation for the optimization of anti-contamination culture medium and the prevention of bacteria or fungi pollution in the tissue culture of aquaticSplants.
