摘    要:微卫星标记由于具有在基因组中分布广、多态性高及共显性等特点,因而在群体遗传多样性分析中广泛应用。本实验针对绵羊19个微卫星标记位点构建了8个多重PCR扩增体系,其中包括3个三重,5个两重PCR扩增体系。采用建立的多重PCR扩增体系对32只小尾寒羊的多态性进行了检测,结果表明19个微卫星标记的等位基因数在5~17之间,期望杂合度、观测杂合度和多态信息含量分别位于0.423~0.885、0.344~0.875和0.392~0.859之间。本实验所建立的多重PCR扩增体系将为绵羊遗传多样性、亲子鉴定和个别识别提供技术基础。

Development and Application of Multiplex-PCR System For Sheep Microsatellite Markers

Abstract:Microsatellite markers have been widely used to study the genetic diversity analysis because of their richness in the genome, their high polymorphism and co-dominance. In the present study, eight multiplex-PCR sets were obtained for 19 microsatellite loci of domestic sheep. Three of them are triplex-PCR sets and five are duplex-PCR sets. Further, genetic polymorphism was examined in 32 small-tailed han sheep by multiplex-PCR system developed in this study. The number of alleles per locus ranged from 5 to 17 based on 32 individuals. The expected and observed heterozygosity and polymorphism information content per locus ranged from 0.423 to 0.885, 0.344 to 0.875, and 0.392 to 0.859, respectively. The results provided a technical basis for the studies of genetic diversity, parentage test and individual identification in sheep.
