| 摘 要： | 为获得墨兰舌瓣总RNA最佳提取方法,以墨兰"南菊"舌瓣为材料,利用CTAB法、华舜植物叶总RNA提取试剂盒、Trizol法、天根RNAsimple总RNA提取试剂盒、Bioteke通用植物总RNA提取试剂盒、Bioteke通用植物总RNA提取试剂盒DNaseI过柱消化法、宝生物RNAiso-mate for Plant Tissue及宝生物RNAiso-mate for Plant Tissue改良法提取舌瓣总RNA。通过RNA产率、纯度、电泳图谱和RT-PCR等分析,确立适用于墨兰舌瓣RNA分离的方法。结果表明:宝生物RNAiso-mate for Plant Tissue改良法提取的RNA除了28S,18S,5SrRNA清晰条带外,RNA的得率和纯度优于其他供试方法。RT-PCR结果进一步证实宝生物RNAiso-mate for Plant Tissue改良法提取的总RNA可用于进一步的分子生物学试验。 |
