| 摘 要： | 以棉籽为原料,探索一种快速、简便的棉籽DNA的提取纯化方法以及PCR检测该棉籽是否为转基因的方法。该方法能从200mg棉籽中提取0.4μg高纯度DNA,用于PCR检测。针对18S rDNA、CaMV35S、NOS、NPTⅡ和CryⅠA(c)等基因进行了棉籽中内外源基因PCR检测,分别扩增出不同的目的片段,为颗粒类产品的外源基因检测提供了可行方法。

| Abstract: | In this paper,a simple and rapid-method which could extracted DNA from cottonseed had been developed.0.4 μg DNA could be obtained from 200 mg cottonseed.PCR amplification of sequences of CaMV35S,NOS,NPTⅡ,CryⅠA(c)and an endogenous reference gene 18S rDNA in refined cottonseed oil was performed.The results showed that the sequence-specific fragments were amplified in the samples,demonstrating that the method could be applied for detection of transgene sequences in refined cottonseed.There are some DNA fragments in refined cottonseed,and PCR amplification can detect the foreign DNA.A feasible method to isolate and detect the foreign DNA in refined cottonseed was provided. |
