金线莲优良种质筛选和快繁技术研究

摘    要:金线莲Anoectochilus roxburgii素有“药王”、“神草”等美称,作为新兴的抗癌中药驰名全国。本实验对不同来源的金线莲进行芦丁、总氨基酸、总有机酸含量进行了检测。结果表明:1)芦丁总平均含量极低,实验组省平均水平为广西(J11)>福建(J1 - J5)>台湾(J6 - J8),又以广西平乐的金线莲(J11)为佳;2)总氨基酸含量栽培种(J1 - J8、J11)均值高于野生种(J9、J10),综合实验组各省表现为台湾(J6 - J8)>福建(J1 - J5)>浙江(J9、J10)>广西(J11),台湾桃园的金线莲(J8)含量明显高于其他几个样品;3)总有机酸含量台湾各来源(J6 - J8)依次高于广西(J11)和福建(J1 - J5),其中台湾桃园的(J8)含量最高。由上述结论得各来源中,台湾桃园的金线莲(J8)种质最为优良。本实验选择该来源种为组织培养材料,摸索其离体培养的最适条件,结果为:含1个茎节长0.8cm,形态学下端长于上端的茎段与1.5mg/L 6-BA + 0.5mg/L NAA的激素配比处理培养基为各自最佳处理条件,两者结合诱导成功率最高,可达91.7%。从而为珍稀药用植物金线莲优良种质的筛选培育和快繁保存提供理论依据。

Studies on germplasm selection andrapid propagation technique of Anoectochilus roxburgiim

Abstract:Anoectochilus roxburgii, is well-known in China as a new anti-cancer traditional Chinese medicine. The contents of rutin, total amino acids and total organic acids in Anoectochilus roxburghii from different sources were detected. The results showed that: 1) the total average content of rutin was very low, and the average level of the experimental group was Guangxi (J11) > Fujian (J1 - J5) > Taiwan (J6 - J8), and the best was Anoectochilus roxburghii (J11) in Pingle, Guangxi. 2)The average content of total amino acids in cultivated species (J1 - J8, J11) was higher than that in wild species (J9, J10), and the performance of Taiwan (J6 - J8) > Fujian (J1) in the comprehensive experimental group was Taiwan (J6 - J8) > Fujian (J1- J5) > Zhejiang (J9, J10) > Guangxi (J11), the content of Anoectochilus roxburghii (J8) in Taoyuan of Taiwan was significantly higher than that of other samples. 3)The total organic acid content of Taiwan (J6-J8) was higher than that of Guangxi (J11) and Fujian (J1-J5), and the content of (J8) in Taiwan peach orchard was the highest. According to the above conclusion, Anoectochilus roxburghii (J8) from Taiwan peach orchard is the best. The results were as follows:the stem segment with a stem node length of 0.8 cm, the lower end of morphology longer than the upper end, and the hormone ratio of 1.5 mg / L 6-BA + 0.5 mg / L NAA was the best treatment conditions. The combination of the two was the highest induction success rate of 91.7%. The results provide a theoretical basis for the selection, cultivation and rapid propagation of Anoectochilus roxburghii.
