木聚糖酶基因xynA在大肠杆菌中的高效表达

摘    要:参照Genebank中疏棉状嗜热丝孢菌木聚糖酶A基因序列,以引物拼接方式,人工合成采用大肠杆菌优势密码子的基因xynA,插入pET-20b载体,获得重组表达载体pET-20b-xynA。重组菌在16℃下诱导效果最好,10 h酶活达到42.05 U/mL,重组酶占全细胞蛋白的47%,粗酶在65℃和pH 5~8条件下很稳定。

High-level Expression of Xylanase A in Escherichia coli

Abstract:According to the gene sequence in Genebank,a codon-optimized xylanase A gene(xynA)from Thermomyces lanuginosus was syn-thesized to construct the expression vector pET-20b-xynA.16 ℃ was the better induction temperature compared with 37 ℃.A maximum activity of 42.05 U/mL was detected after 10 hours of induction,and the expression level of recombinant xylanase was improved to 47%.The rude enzyme was stable at 65 ℃ and over the pH range of 5.0~8.0.
