朱顶红花粉萌发和花粉贮藏性研究

摘    要:采用正交设计筛选适宜朱顶红花粉萌发的最佳培养基,并通过萌发试验了解其贮藏性。研究表明,适宜朱顶红花粉萌发的培养条件为20℃、5%蔗糖、0.01%硼酸、0.05%钙和5%PEG-4000,培养时间为2h。室温贮藏朱顶红花粉1个月之后完全丧失萌发能力,冷藏(4℃)和冷冻(-18℃)有利于保存花粉生活力:短期贮藏(1个月、2个月和3个月),冷藏和冷冻处理的效果由于品种的不同而有变化,长期贮藏(1年)则几乎都表现为冷冻比冷藏具有更高的花粉萌发率。

Abstract:A series of quadrature-designed substrates are introduced in order to select the best conditions for Hippeastrum pollen culture as well as understanding its Preservation characteristic. The result shows that it would be the best for the pollen germination with a culturing condition of 20 ℃, 5% sugar, 0.01% B, 0.05% Ca and 5% PEG-4000. It is also shows that room temperature is not good for pollen preservation of all Hippeastrum species, while both -18 ℃ and 4 ℃ conduce to it. For short term preservation(1 month, 2 months and 3 months), it is hard to say either -18 cc or 4 ℃ is better, because the result varies among different species. But for long time preservation(1 year), it is an overwhelming conclusion that -18 ℃ is better than 4 ℃.
