摘    要:定点单分子DNA芯片有重要用途,但其制备极具挑战性,用光镊、磁镊及原子力显微镜等方法分离单分子DNA,为此建立的平台设备昂贵,且分离效率太低,难以满足实验要求。本研究采用微流控技术,在层流基础上电泳分离与富集单粒微米珠-单根DNA的复合物,旨在为基于微米孔阵列承载这种复合物的定点单分子DNA芯片的制备提供样本。此技术的完善将为单分子DNA芯片制作开辟新的途径。

Abstract:Position-preseted single molecule DNA chips has important applications including sequencing.But it is challenging to fabricate such high density chips.A few methods,such as optical tweezers,magnetic tweezers,and atomic force microscopy(AFM),etc.have been used to separate single DNA molecules.However,they are insufficient for fabrication of high density,single-molecule DNA chips.In this report,a novel microchip has been designed that combine microfluidic laminar flow and capillary electrophoresis to efficiently separate and enrich microbead-single DNA conjugates,each of which could be loaded into an individual well in a microwell array to fabricate position-preseted single-molecule DNA array.
